Embryonic stem cells derived cardiomyocytes are a suitable model for assessment of cardiotoxic effects of doxorubicin and other drugs
Authors
Abstract:
Introduction: Doxorubicin is frequently used for treatment of several types of cancer. Doxorubicin cardiac toxicity has limited the use of this drug. Corticosteroids may prevent doxorubicin induced cardiotoxicity. Therefore the aim of this study was to evaluate mouse embryonic stem cells derived cardiomyocytes as a model to evaluate the effect of Doxorubicin and dexamethasone. Methods: Mouse embryonic stem cells derived cardiomyocytes were treated with different concentration of doxorubicin for 24 hours and the results were compared with control group. In order to exam effect of dexamethasone on cardiotoxicity, mouse embryonic stem cells derived cardiomyocytes were either exposed to 0.1, 1 or 10 µM dexamethasone 24 hours prior exposure to doxorubicin or expose to dexamethasone 24 hours before and during exposure to doxorubicin . Each group were compared with only doxorubicin or dexamethasone treated cells. Results: 5µM doxorubicin was selected as the lowest dose that ceased heart beats in more than 50% of Mouse embryonic stem cells derived cardiomyocytes. Results revealed that 10 µM dexamethasone for 24 hours before treatment with doxorubicin has protective effect on doxorubicin induced cardiotoxicity. Conclusion: The overall results obtained in this model are in accordance with previous literature. Thus suggesting that mouse embryonic stem cells derived cardiomyocytes is a suitable model for assessment of cardio toxic effects of drugs. Key words: Stem cell, Cardiomyocyte, Doxorubicin, Dexamethasone

full textGenetically Engineered Mouse Embryonic Stem Cell – derived Cardiomyocytes as a Suitable Model on Drugs Toxicity In vitro
Background DOX is a powerful chemotherapeutic agent used in the treatment of solid tumors and malignant hematological diseases. However, its cardiac toxicity limits the clinical usefulness of this drug. Previous reports have shown Corticosteroids induce a cytoprotective effect on cardiomyocytes. Mouse transgenic embryonic stem cell-derived pure cardiomyocytes may be considered as a model for a...
full textEvaluation of Chronotropic Properties of Mouse Embryonic Stem Cells-Derived Cardiomyocytes After Fibroblast Growth Factor Treatment
Purpose: We investigated the effect of (bFGF) (basic-Fibroblast Growth Factor) on the differentiation of divided cardiomyocytes from mouse embryonic stem cells (ES) and their pharmacological properties. Materials and Methods: The mouse embryonic stem cells (Royan B1) were cultured as 800 cells per 20µl of a hanging drop. After two days, ES cells in each drop aggregated to form embryoid bodies ...

full textthe effect of doxorubicin on viability and morphology of human embryonic stem cell-derived cardiomyocytes
in spite of serious cardiotoxicity side-effects, doxorubicin is frequently used for treatment of several types of cancers. isolated human adult cardiomyocytes could be the best model for assessing drug-induced cardiotoxicity, while harvesting mature cardiomyocytes is restricted by some limitations such as biopsy size, cell numbers, viability, proliferative capacity and their disability to be pa...
full textThe Effect of Cardio Gel and Matrigel on the Ultrastructure of Cardiomyocytes Derived From Mouse Embryonic Stem Cells
Purpose: To investigate the effect of cardiogel and matrigel on the ultrastructure of embryonic stem cell-derived cardiomyocytes. ECM: Extracellular Matrix derived from cardiac fibroblasts (cardiogel), commercial extracellular matrix (matrigel) and control group (without ECM) were cultured for up to 21 days. Ultrastructural properties of cardiomyocytes were evaluated by transmitting electron mi...
